DAPK
Death associated protein kinase 1 (DAPK) is a serine/threonine kinase involved in the regulation of multiple cellular processes such as apoptosis, autophagy, cell migration, membrane blebbing, etc. Autophosphorylation of DAPK at Ser308 inhibits its catalytic activity and dephosphorylation at this site is an important marker for DAPK activation. The mouse embryonic fibroblasts (MEFs) from DAPK-/- mice showed decreased apoptotic response to overexpression of oncogenes such as c-myc and E2F due to the insufficient activation of the p19ARF-p53 pathway. DAPK is pro-apoptotic in response to both TNF-a and shear stress alone. The dysfunction of DAPK or DAPK related signalling pathways have been linked with various diseases such as cancer, stroke, inflammation and atherosclerosis.
