  • Brief Description
HS-72 is an allosteric, selective, cell-permeable inhibitor of inducible Hsp70 (Hsp70i or Hsp72).
  • Description
HS-72 is an allosteric, selective, cell-permeable inhibitor of inducible Hsp70 (Hsp70i or Hsp72), shows selectivity for Hsp70i over the closely related Hsc70 as well as the broader purinome; inhibits Hsp70i activity in a Huntington’s cell model and across multiple tumorigenic cell lines; causes degradation of HER2 and Akt; is bioavailable in vivo and reduces tumor growth in a spontaneous mouse mammary tumor model; also demonstrates anti-dengue virus activity.

A group of tetratricopeptide repeat (TPR)-containing proteins has been shown to interact with the C-terminal domain of the 70 kDa heat-shock cognate protein (hsc70). In the present study, the effect of the TPR-containing proteins, including the C-terminus of hsc70-interacting protein (CHIP), TPR1 and human glutamine-rich TPR-containing protein (hSGT), on refolding of luciferase by DnaJ and hsc70 was investigated.
