In VitroHBC is a GFP fluorophore-like synthetic dye with a structurally rigid electron acceptor and a strong electron donor, nonfluorescent in solution. ‘Pepper’ is able to enhance HBC’s fluorescence more than 3,000-fold and exhibits bright fluorescence when expressed in living cells.‘Pepper’ has reduced Mg2+ sensitivity after multiple rounds of optimization, displays a well-defined fluorescent profile with an excitation maximum at 485nm, an emission maximum at 530nm, and excellent brightness.HBC514 (2-10 μM) is altered flexibly by simple washing and staining by Pepper based CRISPR imaging in live HeLa cells (expressing sgRNA-Pep1 and dCas9).
