In VitroCORM-3 suppresses caspase-1 activation and the secretion of interleukin (IL)-1β and IL-18 in macrophages in response to lipopolysaccharide (LPS) and ATP. CORM-3 inhibits the oligomerization of the adaptor protein apoptosis-associated speck-like protein containing a caspase recruitment domain (ASC), which is required for NLRP3-dependent caspase-1 activation.

In VivoCORM-3 (4 mg/kg, ip) reduces NLRP3 inflammasome activation and inhibits hyperglycemia-induced inflammation in mice. Animal Model:WT mice (C57BL/6J male, 8e10 weeks old).Dosage:4 mg/kg.Administration:IP for 3 h before i.p. injection of E. coli LPS (10 mg/kg), and IP injection after 7-day-treatment of S0130.Result:Resulted in significantly lower plasma levels of IL-1β and IL-18 in response to LPS challenge in vivo in WT mice relative to that in vehicle-treated control mice, whereas TNF-a levels were unchanged.Had lower expression of cleaved caspase-1 and cleaved IL-1b in response to ATP and nigericin, relative to vehicle control, whereas the expression of procaspase-1 and pro-IL-1β expression was unchanged.
