DescriptionLX-2343 is an non-ATP competitive PI3K inhibitor that negatively regulates AKT/mTOR signaling, thus promoting autophagy and increasing Aβ clearance, also inhibits BACE1 enzymatic activity with IC50 of 11.43 uM; reduced Aβ production involving JNK/APPThr668 pathway inhibition, LX2343 (5-20 uM) dose-dependently decreases Aβ accumulation in HEK293-APPsw and CHO-APP cells, and promotes Aβ clearance in SH-SY5Y cells and primary astrocytes; significantly ameliorates cognitive deficits and markedly ameliorates the Aβ pathology in brains in APP/PS1 transgenic mice.
