DescriptionFLLL-31 is a cell-permeable analog of curcumin that inhibits JAK2-mediated phosphorylation of STAT3 on Tyr705 in cancer cells; induces apoptosis in pancreatic and breast cancer cell lines; also inhibit colony formation in soft agar and cell invasion and exhibits synergy with Doxorubicin against breast cancer cells; decreases STAT3 DNA binding activity and expression, and induces apoptosis in several cancer cell lines; also exhibits anti-inflammatory effects on macrophage activation, lymphocyte differentiation, and pro-inflammatory factor production.

In VitroTetramethylcurcumin (FLLL31; 2.5 and 5 μM;?for 24 hours) downregulats STAT3 phosphorylation and DNA-binding activity in MDA-MB-231 breast and PANC-1 pancreatic cancer cells. Tetramethylcurcumin inhibits cell viability, cell invasion. Tetramethylcurcumin is aeffective inhibitor of STAT3 phosphorylation, DNA-binding activity, and transactivation in vitro, leading to the impediment of multiple oncogenic processes and the induction of apoptosis in pancreatic and breast cancer cell lines.
