DescriptionDiD perchlorate is a lipophilic cyanine dye that can be used in cells tracking.(In Vitro):Two weeks after injection of stained cells, found a single, bright, DiD perchlorate-positive cells located between 15 to 40 μm from the endosteum. Results shows a progressive appearance of cell clusters of decreased dye intensity, which is same for the partitioning of DiD perchlorate label on cell division.(In Vivo):Intra-articular injection of the labeled cells in rats with surgically induced KOA was monitored dynamically by an in vivo imaging system. The red-shifted emission spectra of DiD perchlorate allowed deep-tissue imaging in live animals and the labeling procedure caused no cytotoxic effects or functional damage to haMSCs. This approach is an efficient tracking method for haMSCs in a rat KOA model.
