In VitroGuidelines (Following is our recommended protocol. This protocol only provides a guideline, and should be modified according to your specific needs).Caspase activity assay:1. Incubate the cells according to your normal protocol.2. Wash cells once with PBS and centrifugate.3. Resuspend cells in PBS at a concentration of 1× 107 cells/mL.4. Prepare the standard reaction buffer which includ 100 μM caspase-8 substrate AC-LETD-AFC. Prepare correspounding standard reaction buffer according to your protocol.5. Add 15 ul of the cells suspension to a microplate, and mixed with the appropriate peptide substrate dissolved in a standard reaction buffer.
